THE ROLE OF GENETIC AND ENVIRONMENTAL FACTORS IN AGING AND LONGEVITY

Abstract: 

Background: With an ever-growing proportion of elderly people in Western societies, there is an immense pressure to identify strategies to prevent or delay age-associated frailty and disease that is imperative for maintaining the health of our population, as well as European economy. However, in order to develop therapeutic strategies for delaying age-related pathology, a better understanding of the underlying causes of ageing is required.

Aim: The proposed Program has its core intellectual focus on the causal contribution of Genetic and Environmental Factors on Age-related macromolecular damage and Longevity (GEnAge).

The objectives of the Program are to:

  1. Establish the foundations for systems biology approaches in the field of macromolecular damage in ageing by integrating research from basic mechanisms to translational research applications.
  2. Create a national research network aiming at establishing platforms of excellence for biological tools, technology, training and dissemination that will form the core of the Program.
  3. Transform fruitful, long-term collaborations into a stronger intellectual and training network and build links between the participating labs.

The GEnAge will focus on a series of objectives that are achievable within the time course of the Program and are grouped into three work packages (WP):

  1. The impact of chronic DNA damage in cell fate with advancing age.
  2. The role of protein degradation and proteasomal regulation during the process of ageing.
  3. The role of mitochondrial biogenesis in ageing.

Expected results: The Program is expected to provide a pioneering national framework that will address the basic mechanisms underlying the process of aging, including insights into the natural defense systems that promote longevity as well as providing with gene targets for the development of rationalized intervention strategies against age-related pathology.
